Este trabajo, realizado en el Centro de Biolog\u00eda Molecular Severo Ochoa (<a style=\"color: #000000;\" href=\"https:\/\/www.cbm.uam.es\/es\/\" target=\"_blank\" rel=\"noopener noreferrer\">CBMSO-CSIC-UAM<\/a>) en colaboraci\u00f3n con el Centro Nacional de Investigaciones Oncol\u00f3gicas (<a style=\"color: #000000;\" href=\"https:\/\/www.cnio.es\/\" target=\"_blank\" rel=\"noopener noreferrer\">CNIO<\/a>), e iniciado en el Centro Nacional de Investigaciones Cardiovasculares (<a style=\"color: #000000;\" href=\"https:\/\/www.cnic.es\/\" target=\"_blank\" rel=\"noopener noreferrer\">CNIC<\/a>), ha sido publicado en la revista\u00a0<a style=\"color: #000000;\" href=\"https:\/\/www.cell.com\/cell-reports\/fulltext\/S2211-1247(22)01351-1\" target=\"_blank\" rel=\"noopener noreferrer\">Cell Reports<\/a>.\u00a0<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"color: #000000;\">Las primeras fases de desarrollo transforman el embri\u00f3n reci\u00e9n fecundado, de una sola c\u00e9lula, en un blastocito, que posee unas 60 c\u00e9lulas y presenta dos tipos de poblaciones celulares diferentes: el trofoectodermo, que dar\u00e1 lugar a la placenta, y la masa celular interna, que generar\u00e1 el organismo adulto.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"color: #000000;\">Para descubrir si necesario que el genoma est\u00e9 plegado en tres dimensiones de una manera correcta para que sucedan las primeras fases del desarrollo del embri\u00f3n, los investigadores analizaron los efectos de eliminar desde el principio del proceso un factor denominado CTCF, fundamental para que el ADN se estructure correctamente en el n\u00facleo de la c\u00e9lula.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"color: #000000;\">Mediante modelos gen\u00e9ticos de rat\u00f3n, los investigadores generaron embriones que carec\u00edan de la contribuci\u00f3n materna de ese factor. \u201cEsto era importante, ya que se sabe que, durante la formaci\u00f3n de los gametos femeninos en la ovog\u00e9nesis, la madre deposita tanto prote\u00ednas como ARN mensajeros en el ovocito, que son necesarios para las fases m\u00e1s iniciales del desarrollo tras la fertilizaci\u00f3n. Sin embargo, y para nuestra sorpresa, los embriones modificados se desarrollaban sin problemas e incluso llegaban hasta adultos\u201d, explica el investigador Miguel Manzanares, del Centro de Biolog\u00eda Molecular Severo Ochoa.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"color: #000000;\">Adem\u00e1s, los investigadores observaron que los embriones sin ninguna contribuci\u00f3n de CTCF llegaban sin problemas hasta el estadio de blastocisto, formando correctamente el trofoectodermo y la masa celular interna, y solo a partir de entonces dejaban de crecer y perec\u00edan. Sin embargo, si analizaban la estructura del ADN en el n\u00facleo celular de estos blastocistos, comprobaron que estaba muy afectada y desorganizada.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"color: #000000;\">\u201cNuestro estudio ha revelado que la estructura 3D del genoma no es necesaria para poner en marcha el proceso de desarrollo embrionario, y que, sin embargo, CTCF regula el cambio metab\u00f3lico que tiene lugar durante la transici\u00f3n de masa de c\u00e9lulas surgida de la segmentaci\u00f3n del cigoto a blastocisto necesario para que el embri\u00f3n contin\u00fae su desarrollo\u201d, concluye Manzanares.<\/span><\/p>\n<hr \/>\n<p style=\"text-align: justify;\"><span style=\"color: #000000;\"><strong>Referencia bibliogr\u00e1fica:\u00a0<\/strong><\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"color: #000000;\">Andreu MJ, Albvarez-Franco A, Portela M, Gimenez-Llorente D, Cuadrado A, Badia-Careaga C, Tiana M, Losada A, Manzanares M.
