Pero los cient\u00edficos han dado un gran paso hacia la creaci\u00f3n de c\u00e9lulas de la sangre a partir de c\u00e9lulas madre. Lo han hecho con c\u00e9lulas humanas, aunque el \u00faltimo paso implica trasplantar las c\u00e9lulas precursoras de la sangre (c\u00e9lulas madre hematopoy\u00e9ticas) a la m\u00e9dula \u00f3sea de ratones. El trabajo tiene implicaciones para el tratamiento de la leucemia, y tambi\u00e9n para el cribado de nuevos f\u00e1rmacos.<\/p>\n<p style=\"text-align: justify;\">Dos grupos de cient\u00edficos, del\u00a0<a href=\"http:\/\/www.childrenshospital.org\/\">Hospital Infantil de Boston<\/a>\u00a0y el\u00a0<a href=\"http:\/\/weill.cornell.edu\/\">Weill Cornell Medicine<\/a>\u00a0de Nueva York, han conseguido mediante estrategias distintas derivar de c\u00e9lulas madre un tipo especial de ellas especializado en generar las c\u00e9lulas sangu\u00edneas, desde los gl\u00f3bulos rojos que trasportan el ox\u00edgeno de los pulmones a los tejidos hasta el equipo diverso de linfocitos (o gl\u00f3bulos blancos) que constituyen nuestro sistema inmune. Es un avance que muchos hab\u00edan intentado antes sin \u00e9xito. Y pone a tiro de piedra la creaci\u00f3n de sangre humana al servicio de la medicina.<\/p>\n<p style=\"text-align: justify;\">Los bi\u00f3logos del desarrollo sab\u00edan ya que una sola c\u00e9lula madre hematopoy\u00e9tica (generadora de sangre, literalmente) puede generar toda la notable variedad de linfocitos que discurren por nuestras venas y nos permiten sobrevivir a las infecciones por virus y bacterias, adem\u00e1s de los gl\u00f3bulos rojos. Esas c\u00e9lulas madre, o precursoras de la sangre, residen en la m\u00e9dula \u00f3sea. Es all\u00ed donde se origina todo el contenido celular y la sabidur\u00eda de nuestra sangre. Durante toda la vida. A esto se debe la capacidad curativa de los trasplantes de m\u00e9dula.<\/p>\n<p style=\"text-align: justify;\">\u201cLa capacidad de manufacturar c\u00e9lulas madre hematopoy\u00e9ticas en el laboratorio entra\u00f1a una promesa enorme para la terapia celular\u201d, dicen en\u00a0Nature\u00a0Carolina Guibentif y Berthold G\u00f6ttgens, cient\u00edficos de c\u00e9lulas madre del MRC (Medical Research Council brit\u00e1nico) y la Universidad de Cambridge, no relacionados con el estudio. Estos dos investigadores subrayan tambi\u00e9n los obst\u00e1culos que a\u00fan separan la fabricaci\u00f3n de sangre de la pr\u00e1ctica cl\u00ednica.<\/p>\n<p style=\"text-align: justify;\">En primer lugar, solo uno de los dos trabajos presentados en\u00a0Nature\u00a0(<a href=\"http:\/\/nature.com\/articles\/doi:10.1038\/nature22370\">1<\/a>\u00a0y\u00a0<a href=\"http:\/\/nature.com\/articles\/doi:10.1038\/nature22326\">2<\/a>) aborda una evaluaci\u00f3n del riesgo de que las c\u00e9lulas precursoras de la sangre (hematopoy\u00e9ticas) creadas con estas t\u00e9cnicas puedan generar c\u00e9lulas cancerosas, y solo por un tiempo limitado. Los investigadores han comprobado que sus c\u00e9lulas no generan leucemia (el c\u00e1ncer de las c\u00e9lulas de la sangre) durante las 20 semanas posteriores al trasplante en ratones. Los analistas creen que hay que ampliar ese estudio m\u00e1s tiempo.<\/p>\n<p style=\"text-align: justify;\">En los poqu\u00edsimos casos en que la terapia g\u00e9nica (infectar a un paciente con el gen correcto que le falta), como en el tratamiento de los\u00a0ni\u00f1os burbuja, se ha dado alg\u00fan caso de leucemia inducida por la propia terapia. Para insertar el gen correcto en las c\u00e9lulas del paciente\u00ba, el vector m\u00e1s eficaz hasta la fecha ha sido un retrovirus, un familiar del VIH (el agente del sida) que, como parte de su ciclo vital, se inserta en el genoma de su hu\u00e9sped. Seg\u00fan d\u00f3nde se inserten, estos retrovirus pueden causar la activaci\u00f3n de alg\u00fan gen del c\u00e1ncer, y causar as\u00ed una leucemia.<\/p>\n<p style=\"text-align: justify;\">En cualquier caso, como recuerdan Guibentif y G\u00f6ttgens, las nuevas t\u00e9cnicas de edici\u00f3n gen\u00f3mica, como la flamante CRISPR, pueden lograr insertar los genes correctos sin necesidad de retrovirus, ni de ning\u00fan otro virus. CRISPR hace su trabajo y desaparece despu\u00e9s. Ello, junto a su gran eficacia y facilidad de uso, le ha convertido en la estrella de los laboratorios de gen\u00e9tica de medio mundo.<\/p>\n<p style=\"text-align: justify;\">Quedan, por tanto, problemas t\u00e9cnicos importantes que resolver. Pero ya son solo esos problemas t\u00e9cnicos los que nos separan de la creaci\u00f3n de sangre en el laboratorio.
