Marco-Contelles explic\u00f3 que \u201cen la b\u00fasqueda de nuevos f\u00e1rmacos para su tratamiento, por tratarse de una patolog\u00eda extremadamente compleja y multifactorial, el dise\u00f1o de mol\u00e9culas multipotentes, capaces de actuar simult\u00e1neamente en diversos sistemas enzim\u00e1ticos o receptores implicados en el progreso y desarrollo de la enfermedad, es sin duda una de las estrategias terap\u00e9uticas preferidas, adoptada ya en numerosos laboratorios de investigaci\u00f3n\u201d.<\/p>\n<p style=\"text-align: justify;\">\u201cDe hecho, las hip\u00f3tesis cl\u00e1sicas para tratar la enfermedad de Alzheimer, como la colin\u00e9rgica, del beta-amiloide y de la prote\u00edna tau, no han sido capaces hasta ahora de identificar una mol\u00e9cula eficaz y eficiente, y no lo har\u00e1n posiblemente mientras no se aborde su dise\u00f1o integrando en una sola mol\u00e9cula motivos farmac\u00f3foros, funcionales y estructurales, que sean capaces de desencadenar, simult\u00e1neamente, respuestas positivas en diversas dianas biol\u00f3gicas implicadas en la enfermedad\u201d, a\u00f1ade el investigador.<\/p>\n<p style=\"text-align: justify;\">La pregunta -advierte Marco-Contelles- estriba en decidir cu\u00e1les y cu\u00e1ntas dianas biol\u00f3gicas se van a seleccionar para su dise\u00f1o, partiendo de unas premisas simples y generales, como, por ejemplo, que sea una peque\u00f1a mol\u00e9cula, permeable, antioxidante, con capacidad antagonista de biometales, y neuroprotectora. \u201cEsta pregunta la hemos abordado dise\u00f1ando mol\u00e9culas TRIpotentes, que sean capaces de inhibir las colinesterasas (enzimas que catalizan la hidr\u00f3lisis del neurotransmisor acetilcolina, en la sinapsis, en colina y \u00e1cido ac\u00e9tico), y las monoamino oxidasas A\/B (enzimas que catalizan la oxidaci\u00f3n de monoaminas y la degradaci\u00f3n de neurotransmisores, como serotonina, noradrenalina), y que fueran antagonistas del receptor de histamina H3R, cuya regulaci\u00f3n es bien sabido que permite la liberaci\u00f3n de estos neurotransmisores, tanto en condiciones normales como patol\u00f3gicas, lo que supone que las mol\u00e9culas que act\u00faen sobre H3R pueden tener uso terap\u00e9utico en la enfermedad de Alzheimer.<\/p>\n<p style=\"text-align: justify;\">Esta apuesta original nos ha permitido identificar la mol\u00e9cula MBA354, con el perfil farmacol\u00f3gico in vitro deseado, equilibrado, y con potencias de inhibici\u00f3n o interacci\u00f3n a nivel nanomolar, y como un compuesto-hit, pues, permeable a la barrerahematoencef\u00e1lica, antioxidante y neuroprotector\u201d, explica el investigador. \u201cLa mol\u00e9cula ha mostrado un significativo efecto pro-cognitivo en un modelo in vivo de la enfermedad de Alzheimer, resultado que nos van a permitir ulteriores desarrollos encaminados a encontrar un compuesto-lead para su terapia\u201d, concluye.<\/p>\n<p style=\"text-align:
